چکیده

The fresh water unicellular green alga Chlamydomonas reinhardtii was used to explore whether it could function as a model system to identify proteins that are differentially expressed in response to arsenate exposure. Cells were treated with different concentrations of arsenate ranging from 100 400 M. When exposed to 200 M arsenate, the amount of live cells started to lessen on the second day and continued to diminish, indicating a toxic effect of arsenate. Proteomic analysis was used to investigate if these cells showed a specific response to arsenic-induced stress. Fifteen proteins were found that were over-expressed in the 200 M arsenate-treated samples and two proteins were found to be very strongly over-expressed in samples treated with 400 μM. These were selected for identification using liquid chromatography coupled with tandem mass spectrometry. Oxidative stress and protein damage were the major effects as shown by the up-regulation of Mn-superoxide dismutase, an oxygen-evolving enhancer protein, a chaperonin-like protein and a heat shock protein.

Biosynthesis of Silver Nanoparticles Using Chlamydomonas reinhardtii and its Inhibitory Effect on Growth and Virulence of Listeria monocytogenes

Background: Biosynthesis of nanoparticles using microorganisms, enzymes, and plant extracts is regarded as an alternative to chemical methods. Microalgae appear to be an efficient biological platform for nanoparticle synthesis as they grow rapidly and produce large biomass at lower cost. Objectives: The possibility of silver nanoparticles biosynthesisby freshwater green microalgae, Chlamydomona...

Investigation of an Optimized Context for the Expression of GFP as a Reporter Gene in Chlamydomonas Reinhardtii

Background: Chlamydomonas reinhardtii is a novel recombinant eukaryotic expression system with many advantages including fast growth rate, rapid scalability, absence of human pathogens and the ability to fold and assemble complex proteins accurately, however, obstacle relatively low expression level necessitates optimizing foreign gene expression in this system. The Green Fluorescent Protein (G...

Linking toxicity and adaptive responses across the transcriptome, proteome, and phenotype of Chlamydomonas reinhardtii exposed to silver.

Understanding mechanistic and cellular events underlying a toxicological outcome allows the prediction of impact of environmental stressors to organisms living in different habitats. A systems-based approach aids in characterizing molecular events, and thereby the cellular pathways that have been perturbed. However, mapping only adverse outcomes of a toxicant falls short of describing the stres...

Differential gene expression of Chlamydomonas reinhardtii in response to 2,4,6-trinitrotoluene (TNT) using microarray analysis

The exposure of Chlamydomonas reinhardtii to environmental stress, such as that caused by the explosive 2,4,6-trinitrotoluene (TNT) can alter its gene expression. Expression analysis was conducted using a microarray composed of 3079 Chlamydomonas ESTs to characterize the broad range of responses of gene expression exposed to this common ordnance compound. TNT treatment conditions were determine...

Analysis of motility in multicellular Chlamydomonas reinhardtii evolved under predation

The advent of multicellularity was a watershed event in the history of life, yet the transition from unicellularity to multicellularity is not well understood. Multicellularity opens up opportunities for innovations in intercellular communication, cooperation, and specialization, which can provide selective advantages under certain ecological conditions. The unicellular alga Chlamydomonas reinh...
